Handling
Instructions
Luminex xMAP Carboxylated Microspheres are provided in various
sized containers. The handling of the product varies with the size of
container. Take care to avoid excessive agitation of the product and
resultant foaming/surface precipitation. The microspheres will settle
if left undisturbed. Always ensure that microspheres are
homogeneously resuspended prior to dispensing or analyzing. The
uncoupled microspheres are not monodispersed and tend to
aggregate until coated. Multiple pipetting from the original container
may affect bead concentrations. Protect the microspheres from light
at all times. Follow the resuspension instructions based on the size of
container in use:
For 1 mL containers: Remove from 2-8°C and allow equilibration to
room temperature. Centrifuge for 1-2 minutes at 10,000 x g. Sonicate
in a water bath sonicator to disperse the microsphere pellet. Mix by
gentle vortexing immediately prior to dispensing. Inversion is not
recommended.
For larger containers: Remove from 2-8°C and allow equilibration to
room temperature. Place on rotator and rotate for a minimum of 15
minutes. Mix by gentle inversion immediately prior to dispensing.
For these containers, vortexing and rocking are not recommended.
Bead Selection You select beads from the standard 100-region bead map. Refer to
Table 1. Luminex recommends that you select beads starting from
the upper left, reading across to the end of a row, and then down to
the next row:
