SECTION 1 General Information 1.1 Introduction The Horizontal Midi-Gel systems, (GC)MGU-402T, (GC)MGU-502T, and (GC)GCMGU602T, provide a compact format to rapidly separate and identify DNA and RNA fragments, PCR products and synthetic oligonucleotides, while using minimal quantities of expensive reagents. The tapered baffle system allows gel casting directly onto the unit or removable trays without having to seal with agarose plugs or tape. The gel bed and gel tray are UV transparent.
SECTION 2 Description of Parts 2.1 Unpacking Please verify that your Midi-Horizontal Kit comes complete with the following components: Standard Midi-Horizontal Kit MGU-402T, MGU-502T, MGU-602T Complete Package: • Electrophoresis unit with tapered baffles and UV transparent gel bed • 1 UV transparent gel tray with 2 or 4 comb position slots • 1 adjustable height comb • Safety cover and power leads • Platinum electrodes 2.
SECTION 3 Instructions for Use 3.1 Midi-Gel Unit Preparation and Gel Casting 1. Place Midi-Horizontal unit on a level work surface in an authorized work area. Place the adjustable height comb assembly over the gel bed or gel tray. Loosen the white screws holding the comb to the backing and adjust comb depth (generally 1mm). Gently tighten the adjusting screws and set the comb aside. See Tables on pages 11-12 for comb volumes. 2.
3.3 Gel Casting in Unit using tapered baffles 1. Remove lid by pushing down on white thumb posts while pulling up lid with fingers. DO NOT PULL ON POWER CORDS! 2. Place gel tray in center of horizontal gel bed. 3. Remove tapered baffles from storage position and slide them snugly into the baffle guides next to the bed, handles to the outside. The tapered baffles will rest friction tight against the gel bed. Do NOT press the baffles too tightly into the stops. 4.
6. Remove comb(s). 7. Remove baffles from their casting position. 8.Turn baffles 180º (handles facing in) and place into the baffle storage slots provided at either end of the unit near both terminals. Go to Section 3.5 3.4 Gel Casting using External Gel Caster 1. Level external gel caster using white thumb screws on base. Place gel tray into External Gel Caster. Ends of gel tray friction fit onto place. Position the comb into receptacle slots of gel tray. Add additional comb if desired. 2.
3.5 Running the Gel 1. Add enough running buffer to fill both reservoirs and overflow the surface of the gel to a depth of 2-3mm. Flush out any air bubbles in the wells. Stains may be added during this step. Use according to manufacturer’s recommendations. 2. Load the samples into sample wells. Do not forget to load DNA size standard ladders. For best visibility load samples on contrasting background color such as red or black. Safe stains may be added during this step.

SECTION 4 Running Conditions 4.1 4.2 Recommended Power The recommended power conditions for optimal resolution are 50 to 150V, constant, and 25 to 80 mA. The usual run time will vary for the voltage chosen but should range from 15 to 60 minutes, monitoring nucleic acid migration by progress of marker dyes. Constant power is not a necessity, but it produces uniform heat throughout the run. Be sure the polarity is correct i.e.
SECTION 5 Maintenance of Equipment 5.1 Care and Handling The plastic components of the Midi-Horizontal units are fabricated from acrylic and polycarbonate. Electrodes and connectors are made from pure platinum, stainless steel, and chrome plated brass. As with any laboratory instrument, adequate care ensures consistent and reliable performance. After each use, rinse buffer chamber, gel tray and combs with de-ionized water. Wipe dry with a soft cloth or paper towel, or allow to air dry.
