3.2 Gel Casting in Unit using tapered bafes
1. Liftlidsothatitrestsinupposition.
2.Placegeltrayincenterofhorizontalgel
bed.
3. Removeblacktaperedbafesfromstor-
agepositionandSlidethemsnuglyinto
thegroovesnexttothebed,handles to
the outside.Thetaperedbafeswillrest
frictiontightagainstthegelbed.DoNOT
pressthebafestootightlyintothestops.
4. Positionthegelcomb,remembering,cur-
renttravelsfromnegative(blackcathode)
towardspositive(redanode).Addaddi-
tionalcombsifdesired.
5.Makeagarosesolutionaccordingtomanu-
facturer’sinstructions.Use caution when
handling hot agarose solution! Pourmol-
tenagarosesolutionintogeltray.Allowto
cooluncovereduntilagaroseturnopaque.
Stains may be added during this step. Use
according to manufacturer’s recommenda-
tions.
