9.Removeentireassemblyfrompyrexdishandloadintodesiredposition(slot)inbufferchamberwithblack
screenfacingtowardsblackterminal(cathode)andredscreenfacingtowardsredterminal(anode).
3.3 STANDARD ELECTRO-BLOT TRANSFER
1. Fillthechamberwithtransferbuffer.(Buffersshouldbepreparedfreshwithreagentgradechemicalsand
pre-cooled).Thetransferbuffershouldcomeuptothetopoftheplatinumlabyrinth.Thebuffershouldnot
comeincontactwiththebananaplugswhenthegelcassettesandwich(es)areimmersedintheunit.
2. Align safety cover over the unit and carefully attach. Begin cycling of coolant.
3. Connecttheleadstothepowersupply,matchingthecolor-codedredtoredandblacktoblack.SeeSection
4.1forrecommendedpowerconditions.
4. Transfertimeswillvaryaccordingtoseveralparameters.Optimizationofelectro-blottingtransfersmustbe
determinedempirically.Keepinmindthefollowingprinciplesthatgovernthemovementofmoleculesofgel
electrophoresis:
• Thickerorhigherpercentagegelswilltakelongertotransferthanthinnerorlowerpercentagegels.
• Largemoleculeswillneedextendedtransfertimestocompletelytransfer.
• Actualtransfertimesfordenedconditionscanbeapproximatedbyrunningmolecularweightstandards.
3.4 REMOVING THE CASSETTES
1. Turnthepowersupplyoffanddisconnecttheleadsfromthepowersupply.
2. Removethesafetycoverfromtheunit,byplacingthumbsonwhitepostsnexttored&blackconnectors,
thenpushingdownwhilepullingupwithngersunderlid.DONOTpullonpowercords.
3. Gentlyliftthecassettefromtheunit.Alwaysweargloves,eyeprotectionandprotectiveclothingifbufferand/
orgelcontainEthidiumBromide.EthidiumBromideisapowerfulmutagen;gloves,eyeprotectionandpro-
tective clothing should always be worn when handling the gel or buffer solutions. See Material Data Safety
Sheets.
4. Marktheorientationofthemembranewithapencilorbycuttingoffacornerandtakeapartthecassette
carefully.
5. Processmembraneaccordingtotypeoftransferandmanufacturer’srecommendations.
