7.4 Sample Preparation
1. Determine the appropriate concentration of sample to load (depends on the load volume and the
detection method used).
2. Dilute the sample with at least an equivalent volume of sample buffer.
For example, combine: 5 μl sample
5 μl sample buffer
10 μl total volume
7.5 Running Conditions
Power conditions (stepwise) 100 V constant 60 min
250 V constant 60 min
500 V constant 30 min
Starting current 5–25 mA/gel
Final current 5–25 mA/gel
Run time 150 min
