Issue:IntheRBSAnalysistool,iftheuseropensacompletedjobandre‐runstheanalysis,theoriginal
jobwillshowa‘Running’statusifyoudoubleclickthisjob.
Workaround:Whenthesecondjobiscompleted,thestatusoftheoriginaljobwillreverttothecorrect
statusof‘Completed’.
Multiple parts with a name containing special characters do not get permuted correctly
Issue:SinceAutoDesigndoesnotacceptspecialcharactersinaPartname,theAutoDesigntooldrops
thespecialcharactersandincludesonlythealpha‐numericcharactersinthePartname.
IncaseofmultiplePartshavingspecialandalpha‐numericcharactersinthePartname,inAutoDesign,
whenthespecialcharactersareexcluded,oneormorePartscouldincludethesamealpha‐numeric
charactersintheexactsamesequenceasoneanother,andthereforeonlyonePartwillbeincluded.
Workaround:DonotincludespecialcharactersinthePartnames.
Software becomes unresponsive on generating a Dot Plot for a large sequence

Analysis>GenerateDotPlotfunctionforalargesequence.
Workaround:Refrainfromperformingthisfunctionifthesequenceislargerthan1500bp.
Duplicate warning messages appear when invoking Assembly Compatibility Tool
Issue:Onsomeoccasions,duplicatewarningmessagesappearwithintheCompatibilityCheckErrors/
WarningstablewhileperformingcloningusingeithertheSeamless/HighOrder(Liang,2012),SLIC(Li,
2007),CPEC(Quan,2009)andExo/FillIn/Ligation(Gibson,2009)assemblymethods.
Workaround:Theduplicatewarningmessagesarethesame.Clickingoneitherofthemessageswill
bringupthedialogboxforthecorrectionaction.
Same sequence is returned as the terminator sequence for very short sequences
Issue:ThesamesequenceisbeingcalculatedastheterminatorsequencewhenrunningtheTerminator
Calculatortoolagainstveryshortsequences(<10bps).
Workaround:OpenuptheRNAStructuretool,andmanuallycheckthesecondarystructuretoexamine
theterminatorsequence.
An additional warning message for checking non-adjacent homologous sequence appears in the
Compatibility Check Errors table.
Issue:DuplicatewarningmessagesappearwithintheCompatibilityCheckErrorstablewhilechecking
non‐adjacenthomologoussequences.
